Alesta Therapeutics is a Leiden-based biotechnology company founded in 2021 that develops orally active small molecules for rare genetic diseases. Its lead programme, ALE1, is a potential first oral therapy for hypophosphatasia, a rare bone disease caused by mutations in the ALPL gene; a second programme, ALE2, targets Charcot-Marie-Tooth disease caused by tRNA mutations. BioMarin Pharmaceutical agreed to acquire the company in August 2026 for $275 million upfront and up to $215 million in development and regulatory milestones.
